EFFECTS OF GLUCOSE AND INSULIN ON INTEGRIN EXPRESSION IN RETINAL MICROVASCULAR CELLS.

An increased expression of avb3 and avb5 integrins is associated to angiogenesis in proliferative diabetic retinopathy. In the present study, we examined the effect of high glucose concentrations and insulin on the expression of these two integrins in retinal capillary cells

Endothelial cells (BREC) and pericytes (BRP) from bovine retinal microvessels were incubated in the presence of either glucose (5 mM and 30 mM), 30 mM mannitol, insulin (200 ľU/ml) or insulin + glucose (200 ľU/ml and 30 mM respectively). Expression of avb3 and avb5 integrins was analyzed by capture ELISA. In basal conditions (5 mM glucose), these two integrins were more expressed in BRP than in BREC. When cells were cultured in the presence of 30 mM glucose, integrin expression was not modified in both cell types. On the contrary, insulin alone had opposite effects on BREC and BRP. Indeed, in BREC integrin expression was increased whereas in BRP it was decreased. In the presence of insulin + glucose no differences were observed on BRP integrin expression when compared to insulin alone, whereas different results were obtained with BREC.

These observations suggest that, in retinal capillaries, insulin alone or in association with high glucose concentrations could favour angiogenesis increasing BREC integrin expression. Insulin could also contribute to the pericyte loss observed in diabetic retinopathy by alteration of cell-matrix interactions mediated by pericyte integrins avb3 and avb5.
